Potassium Dihydrogen Orthophosphate Anhydrous KH2PO4

Di-potassium Hydrogen Orthophosphate Tryhydrate
K2HPO4.3H2O 90 g
Potassium Chloride KCL 10 g
Magnesium Sulphate MgSO4.7H2O 10 g
Dissolve in 800 ml Distilled H2O then bring to 1 litre with
distilled H2O
TRACE ELEMENT SOLUTION - 1 LITRE
Sodium Tetraborate 0.04 g
Copper Sulphate 0.4 g
Ferric Orthophosphate 0.8 g
Manganese Sulphate 0.8 g
Sodium Molybdate 0.8 g
Zinc Sulphate 8.0 g
Dissolve Ferric Orthophosphate in 100ml Distilled H2O + 20 ml 4N HCl
- then add the rest of the elements one by one.
Bring volume to 1 litre.
LIQUID MINIMAL MEDIUM -N - 1 LITRE
Glucose 10 g
Salts Solution 10 ml
Trace Elements 1 ml
Distilled H2O to 1 litre.
Dispense 200 ml into 1 litre flasks (or bottles if flasks not available).
Sterilise 10 lbs for 15 min.
(A slight precipitate may develop, but can be ignored)
ASPERGILLUS BUFFERED MINIMAL (M3) - 1 LITRE
Glucose 10 g
Salts Solution 10 ml
Trace Elements 1 ml
Sodium Nitrate 2 g
Agar 12 g
Dissolve in 700ml distilled H2O by heating, bring volume to 1 litre
with distilled water and dispense into 200 ml bottles.
Sterilise 15 lbs for 15 min.
(A slight precipitate may develop, but can be ignored)
ASPERGILLUS M3-CN - 1 LITRE (For addition of alternative C- or
N-sources)
As M3, but omit glucose and sodium nitrate
SOS-N - 1 LITRE (For protoplast regeneration, allowing for selection on
alternative N-sources)
As M3, but
Sucrose 342 g (in place of glucose)
Omit sodium nitrate
Difco agar 20 g (in place of crude agar)
LIQUID COMPLETE MEDIUM - 1 LITRE
Salts solution 10 ml
Glucose 10 g
Sodium nitrate 6 g
Difco Bacto Peptone 2 g
Yeastrel (yeast extract) 1 g
Casein Hydrolysate 1.5 g
Trace Element Solution 1 ml
Vitamin Solution 1 ml
Dissolve in 700 ml, then make up to 1 litre
Autoclave 10 lb for 15 mins.
COMPLETE MEDIUM - 1 LITRE
As Liquid Complete Medium, but add
Agar 12 g
VITAMIN SOLUTION - 100 ml
Riboflavin 0.1 g
Nicotinamide 0.1 g
p-Amino Benzoic Acid 0.01 g
Pyridoxine HCl 0.05 g
Aneurine HCl 0.05 g
Biotin 1.0 mg
Distilled H2O to 100 ml
