cultural_requirements: For biosynthetic pathway, see Fig. 15. Leucine mutants have been used extensively for studies of regulation (see references 427 and 833). Leucine mutants acquire suppressors when grown on Difco agar-sorbose medium. The suppressors are leaky auxotrophs blocked at various steps in sulfur metabolism; apparently these blocks allow more efficient use of the traces of leucine in the agar (425; S. R. Gross, personal communication). Most aliphatic and aromatic amino acids can inhibit growth of leucine mutants at appropriate concentrations, probably because of competition for a common uptake system (S. R.
